一 TBE300A高速逆流色谱(HSCCC)特点:

  • Abstract: A preparative high-speed counter-current chromatography (HSCCC) was used to isolate and separate curculigoside and curculigoside B from the famous traditional Chinese medicinal herb Curculigo orchioides. Some parameters including two-phase solvent system, separation temperature,flow rate of the mobile phase and revolution speed of the apparatus were all investigated, and a successfully isolation and purification was achieved at the following conditions: the two-phase solvent system composed of ethyl acetate-ethanol-water at a volume ratio of 5:1:5(v/v/v) was selected and the lower phase of the system was used as the mobile phase at the flow rate of 2.0ml/min, and the isolation temperature and revolution speed were set at 30◦C and 800rpm.The isolation produced a total of 14.5 mg curculigoside B and 72.8 mg curculigoside with purities of 96.5% and 99.4% determined by high performance liquid chromatography (HPLC) from 300 mg crude extract after cleaning-up by D101 macroporous resin,which was necessary for the excellent purification. The recoveries of the two compounds were 91.6% and 92.5%, respectively, and the chemical structure identification was carried out by UV, MS and NMR. This separation method was more effective than some conventional techniques.

  • 摘 要: 采用高速逆流色谱法分离纯化丹参水溶性成分丹酚酸类物质,制备丹酚酸B化学对照品。分离采用的溶剂系统为正己烷2乙酸乙酯2水2甲醇(1.5:5:5:1.5),上相做固定相,下相做流动相,流速为1.7mL/min,仪器转速850rpm,进样量80mg,纯度用HPLC 方法测定。结果表明:一次分离可制备63.4mg丹酚酸B,其纯度为98.6%。该方法操作简单,可作为高纯度丹酚酸B化学对照品的制备分离方法。

  • Abstract:Pseudostellarin B (cyclic peptide) was isolated and purified from the herbs of Pseudostellaria heterophylla (Miq.) Pax for the first time by high-speed counter-current chromatography (HSCCC) using a two-phase solvent system consisting of n-butanol–ethyl acetate–water (0.6:4.4:5,v/v).The technique can isolate mg levels of the target compound per run with purity better than 96%. The chemical structure of the compound has been positively confirmed by electrospray ionization time of flight (TOF) MS,H-NMR,C-NMR and H-C COSY analyses.

  • Abstract: High-speed counter-current chromatography (HSCCC) technique in semi-preparative scale has been successfully applied to the separation of bioactive flavonoid compounds, liquiritigenin and isoliquiritigenin in one step from the crude extract of Glycyrrhiza uralensis Risch. The HSCCC was performed using a two-phase solvent system composed of n-hexane–ethyl acetate–methanol–acetonitrile–water (2:2:1:0.6:2,v/v). Yields of liquiritigenin (98.9% purity) and isoliquiritigenin (98.3% purity) obtained were 0.52% and 0.32%. Chemical structures of the purified liquiritigenin and isoliquiritigenin were identified by electrospray ionization–MS (ESI–MS) and NMR analysis.

  • Abstract:A preparative high-speed counter-current chromatography (HSCCC) with a two-phase solvent system composed of ethyl acetate–nbutanol–water (2:7:9, v/v/v) was successfully performed to isolate and separate clemastanin B and indigoticoside A from the plant of Radix Isatidis, a traditional Chinese medicine. A total of 59.2 mg clemastanin B and 66.1 mg indigoticoside A with purities of 94.6% and 99.0% determined by high performance liquid chromatography (HPLC) were obtained in one-step elution from 250 mg crude extract, which contained clemastanin B 24.8% and indigoticoside A 28.4%, and the recoveries of clemastanin B and indigoticoside A were 90.3% and 92.2%,respectively. The chemical structure was identified by IR, MS, H-NMR and C-NMR.

  • Abstract:Three flavonoid glycosides including orientin, vitexin, quercetin-3-O-neohesperidoside and one unknown compound were isolated and purified by high-speed counter-current chromatography (HSCCC) and semi-preparative HPLC from Trollius ledebouri Reichb.,a traditional Chinese medicine. Preparative HSCCC with a two-phase solvent system composed of ethyl acetate–n-butanol–water (2:1:3, v/v/v) was successfully performed by increasing the flow-rate of the mobile phase from 1.5 to 2.5 ml/min after 190min. Consequently, 95.8mg orientin,11.6mg vitexin, 9.3mg unknown compound with purities of over 97% and one partially purified peak fraction (contained quercetin-3-Oneohesperidoside at 85.1% purity)were obtained from 500mg of the crude extract. Then the partially purified fraction was further purified by reversed-phase semi-preparative high-performance liquid chromatography. The structure identification of all pure fractions was carried out by UV, MS, 1H NMR and 13C NMR.
